Cell Culture Supplements

Recombinant Human Serum Albumin (rHSA), Animal-free has been produced from a plant based (rice) expression system. rHSA has the identical amino acid sequence as the major HAS isoform found in human serum. Albumin is a cell culture supplement that functions as a carrier protein for fatty acids, growth factors and trace minerals. Albumin also protects cells from toxins and physical shear. Albumin is used to reduce or eliminate the need for serum and to maximize cell growth, viability and productivity in classical, commercial and custom media.

Optimum concentration

The optimum concentration of albumin for cell culture applications varies with the type of cell and the composition of the medium. Albumin is typically used concentrations of 0.5 to 2 g/L in media formulations.

Procedure

Prepare concentrated stock solution of rHSA by dissolving in cell culture grade PBS or water. Use gentle mixing or inversion to minimize the formation of bubbles. Allow rHSA to dissolve for at least 1 hr. Liquid stock should be sterile filtered prior to use. We recommends a pre-filtration step with a 0.8 um filter prior to a final sterile filtration step with a 0.45 µm or 0.2 µm filter. We recommend the adaptation of cells to rHSA supplemented medium for a couple of passages to insure optimum performance. Following the adaptation, the end user can determine the optimum concentration of rHSA for their particular purpose. Cells previously adapted to a medium containing albumin typically require little adaptation to rHSA.
