2X Sample Buffer with Reducing Agent

62.5 mM Tris-HCl (pH 6.8), 2% SDS, 25% (v/v) glycerol, 0.01% bromophenol blue, 5% β-mercaptoethanol or 100 mM DTT (added fresh)

10X Tris-Glycine Running Buffer

Dissolve 30.0 g Tris base, 144.0 g Glycine and 10.0 g SDS in 1000 ml of ddH2O.

10X MOPS Running Buffer

Dissolve 60.6 g Tris base, 104.6 g MOPS, 10.0 g SDS and 3.0 g EDTA in 1000 ml of ddH2O.

10X MES Running Buffer

Dissolve 60.6 g Tris base, 97.6 g MES, 10.0 g SDS and 3.0 g EDTA in 1000 ml of ddH2O.

1X Running Buffer

Dilute 100 ml 10X running buffer with 900 ml ddH2O.

10X Transfer Buffer

Dissolve 30.0 g Tris base and 144.0 g Glycine in 1000 ml of ddH2O.

1X Transfer Buffer

Cool 1X transfer buffer to 4°C before using.

Dilute 100 ml 10X transfer buffer with 200 ml methanol and 700 ml ddH2O.

Note: For the proper high molecular weight proteins, up to 0.1% SDS may need to be added to 1X transfer buffer.

<CosmoPAGE Tris-Glycine Running Buffer>

Description

CosmoPAGE Tris-Glycine Running Buffer Powder is used for electrophoresis on CosmoPAGE Tris-Glycine Precast Gel or Laemmli Tris-HCl gels in Tris-Glycine buffer system. It is convenient and universal for electrophoresis in Tris-Glycine buffer system.

Preparation for use

Reconstitute 1 pouch of buffer powder with 1000 mL deionized water to make 1X running buffer. The pH of the buffer will be around 8.5 and no pH adjustment is required.

Applications

CosmoPAGE Tris-Glycine Running Buffer has been used as a running buffer in sodium dodecyl sulfatepolyacryamide gel electrophoresis (SDS-PAGE). It is usually used for both the anode buffer and the cathode buffer.

<CosmoPAGE MOPS Running Buffer>

Description

CosmoPAGE MOPS Running Buffer Powder is used for electrophoresis on CosmoPAGE Bis-Tris Precast Gel or polyacrylamide gels in Bis-Tris buffer system. It is convenient and universal for electrophoresis in Bis-Tris buffer system.

Preparation for use

Reconstitute 1 pouch of buffer powder with 1000 mL deionized water to make 1X running buffer. The pH of the buffer will be around 7.7 and no pH adjustment is required.

Applications

CosmoPAGE MOPS Running Buffer has been used as a running buffer for sodium dodecyl sulfatepolyacryamide gel electrophoresis (SDS-PAGE) in Bis-Tris buffer system. As compared with MES buffer, this buffer is suitable for separating large and medium-sized proteins.

Tips for using CosmoPAGE

	1. Before sample loading, rinse the wells to remove storage buffer.
	2. Ensure tight-sealing of inner chamber when adaption (or fill outer chamber with buffer to the highest level.)
	3. Under water flow, insertgel remover between gel and cassette from well site of gel. Slowly push gel remover to the bottom of gel until gel is detached from cassette. * Avoid diagonally peeling the gel from the corner or peeling from the bottm.

Tips for Western transfer

	1. Assemble transfer sandwich by orientating cathode, sponge, 3 filter papers, gel, membrane, 3 filter papers, sponge, and anode. (The protein goes to the direction of cathode to anode.)
	2. Remove air bubbles or excess buffer between gel and membrane by carefully moving roller over the membrane.
	3. Maintain whole transfer process at low temperature. (Keep transfer tank in cold toom or on ice.)
